How many second stringers do people normally select for a friendly ? I've thrown in quite a few 17/18 year olds in and I'm using 5-4-1 for only the second time.
I've already got 4-5-1 up to excellent and I'll use this with pressing in most away games and 4-4-2 at home. A lot of clubs seem to use 3-5-2 but if I'm training defenders I'm not so sure this is the right move for me.

All are usually second stringers unless I have too few in which case my worst first teamers play
Reasons:
i) I don't want my best players to get injured unnecessarily
ii) The more players you use, the more players get training even if it is only a change in form

Oh and 3-5-2 is definitely not the best if you're training defending.
3-5-2 is just generally seen as a good formation for Div VI as it helps midfield get better.
As BA Says, the more different players you can play, the better for your team. if your training defence I would play 5-4-1 in friendlies and either the same or 4-5-1 in league games otherwise you may as well train something else. Defensive training takes the longest so you have to make sure you're training as many players as possible, I'm playing 5 at the back in both league and friendlies so I get 10 training slots per week.
5 passable YP's line up against BRB tomorrow, hoping for at least a couple of pops.
